Summary Mesophyll protoplasts of wild pear (Pyrus communis var. pyraster L., Pomoideae) were chemically fused with cell suspension protoplasts of cherry rootstock Colt (Prunus avium x pseudocerasus, Prunoideae), following an electroporation treatment of the separate parental protoplast systems. Fusion-treated protoplasts were cultured, on modified K8P medium, where it had been previously established that neither parental protoplasts were capable of division. Somatic hybrid calli were recovered and, following caulogenesis on MS medium with zeatin and after rooting of regenerated shoots, complete trees were obtained and grown in vivo. Hybridity of these trees was confirmed based on morphological characters, chromosome complement and isozyme analysis. Two separate cloned lines of this intersubfamilial rootstock somatic hybrid (wild pear (+) Colt cherry) were produced. This is the first report of the production of somatic hybrid plants of two woody species, of agronomic value, within the order Rosales. 相似文献

Extinction of small, closed populations in captivity as well as in the wild is believed to be nearly inevitable, because inbreeding
will adversely effect reproductive success, mortality, sex ratio and also the susceptibility to epidemic diseases and environmental
stress. An ever increasing number of primate species exist only in small isolated populations, which contain only a part of
the original genetic variability. In captive breeding programs research about genetic management strategies is, therefore,
of essential importance. In 1980 we imported 9Loris tardigrdus nordicus (4 females, 5 males) from NE-Sri Lanka. The founders came from one natural breeding population. All sexual mature females
are breeding. Up to now the colony contains 36 living individuals. The main goal of our long-term genetic management plan
was to minimize inbreeding and to preserve the genetic diversity. Therefore, we try to pass the founder bottleneck rapidly
by enlarging the population to a desired minimum population size of 25 pairs and to equalize the founder representation within
any generation. The need to control the spread of sublethal genes, introduced by one of the founders, conflicts directly with
the aim of equalizing the founder representation. A solution of this problem is discussed. To produce a sufficiently large
population we intend to give animals to other institutions and to build up an exchange-system for offspring individuals, which

A series of coal mine spoils (5, 10, 12, 16 and 20-yr old) in a dry tropical environment was sampled to assess the changes with time in spoil characteristics, species composition and plant biomass. Coarse fragments (>2 mm) decreased with age of mine spoil while the proportion of 0.2–0.1 mm particles increased. Total soil N, mineral N, NaHCO3-extractable Pi, and exchangeable K increased with age of mine spoil and these parameters were lower in mine spoils than native forest soil even after 20 years of succession. Exchangeable Na decreased with age of mine spoil and in 20-yr old spoil it was higher than native forest soil. Plant community composition changed with age. Only a few species participated in community formation. Species richness increased with age, while evenness and species diversity declined from 5-yr old to 16-yr old community with an increase in the 20-yr old community. A reverse trend occurred for concentration of dominance. Area-weighted shoot and root biomass of other species increased with the age of the mine spoil while that of Xanthium strumarium patches declined with age. Data collected on spoil features, microbial C, N and P, and shoot and root biomass when subjected to Discriminant Analysis indicated a continued profound effect of age. 10 and 12-yr old mine spoils were closer to each other, and 5 and 20-yr old spoils were farthest apart. 相似文献

Life history parameters including longevity, developmental time, and reproduction were determined for whiteflies, Trialeurodes vaporariorum (Westwood), kept in clip-on cages on susceptible parent, Lycopersicon esculentum, resistant parent, L. hirsutum glabratum, and intermediate breeding lines of tomato. Using the Lewontin triangular reproductive function, the intrinsic rate of increase (rm) was calculated for each genotype. This is an elegant tool for detecting resistance, as it incorporates all salient factors of life history. Comparing rm to the other parameters measured, selection criteria were chosen. Developmental time is not a suitable selection criterion as it varies very little even between the most susceptible and the most resistant genotypes. Additionally, developmental time of T. vaporariorum offspring was found to be highly significantly correlated to parental age on all tomato genotypes except L. hirsutum glabratum. Total reproduction, truncated population counts, and longevity appear good criteria for selection. This test, focusing on antibiotic factors, shows large differences between the resistant and susceptible parent genotypes, but much smaller differences between the breeding lines and the susceptible parent. Earlier screenings relying on a variety of tests appear to have maintained antixenotic rather than antibiotic properties in the breeding lines. 相似文献

An increasing world population and rise in demand for tree products, especially wood, has increased the need to produce more timber through planting more forest with improved quality stock. Superior trees are likely to arise from several sources. Firstly, forest trees can be selected from wild populations and cloned using macropropagation techniques already being investigated for fruit tree rootstocks. Alternatively, propagation might be brought aboutin vitro through micropropagation or sustained somatic embryogenesis, with encapsulation of the somatic embryos to form artificial seeds. Tree quality could be improved through increased plant breeding and it is likely that experienced gained, to date, in the breeding of fruit species will be useful in devising strategies for forest trees. Since the development of techniques to regenerate woody plants from explant tissues, cells and protoplasts, it is now feasible to test the use of tissue culture methods to bring about improvements in tree quality. Success has already been achieved for tree species in the generation of somaclonal and protoclonal variation, the formation of haploids, triploids and polyploids, somatic hybrids and cybrids and the introduction of foreign DNA through transformation. This review summarizes the advances made so far in tree biotechnology, and suggests some of the directions that it might take in the future. 相似文献

榆中贝母(Fritillaria yuzhongensis)生长于马啣山的高山地带,全生育期约100 d左右,是一种生育期较短的类短命植物。在果实成熟时,种子的胚发育不全,具有后熟作用的特点,包括形态后熟和生理后热两个阶段;因此,需要一定时期的低温处理,才能完成后熟过程,以解除休眠,促使种子发芽。本试验表明:榆中贝母完成形态后熟过程的最佳温度是7℃左右,经过100d以上的低温处理才能使原胚发育成熟,继续在7℃低温下处理70d以上,才能完成生理后熟过程。榆中贝母种子休眠时间长,需要经过170d以上的低温处理,种子才能萌发。本试验结果为榆中贝母种子的萌发提出了适宜的温度范围和低温处理时间,为提高种子的出苗率和缩短栽培年限提供了科学依据。 相似文献

禾本科植物是粮食作物的主要来源,随着人口的增加和生活水准的提高,人类对粮食的产量、种类和质量的需求也日益迫切。根据国外在1982年对90个发展中国家的统计和预测的结果说明到1990年末,这些国家共缺少72百万吨谷物而到2000年将缺少144百万吨谷物。近十余年以来,随着植物分子生物学的迅猛发展和作为植物生物技术重要组成部分的植物组织培养技术的日臻完善,被公认为非常困难从事的禾本科植物(Gramina-ceae)的组织培养也取得了异常迅速的发展,并且已经在作物改良的生产中取得了成效,显示了越来越大的潜能和威力,为人类从根本上解决食物问题指出了一条切实可行的途径。本文拟在评述近年来禾本利植物组织培养(主要指胚胎培养、器官培养、细胞培养和原生质体培养)的理论性研究和应用性研究的进展,并重点描述和讨沦在应用上较为成熟和有发展前景的几个领域的发展现状以及利用禾本科植物的组织培养技术而进行的基因转移技术的概况。希望能为我国从事植物组织培养的工作者们提供某些参考资料并对于一些问题进行共同的商榷和探讨。 相似文献

对武汉东湖三个定点观测站和潜江市一深水机井中异养细菌群落的种群组成、对不同基质的反应、菌株和群落的生理特征和活力水平等方面比较研究结果表明:环境差异较小的东湖Ⅰ、Ⅱ、Ⅲ站异养细菌群落间的差异亦较小,具有类似的结构特征和生理活力水平,对特定基质表现了相似的作用模式;地下水中的异养细菌群落在上述几方面均显示了明显的差别。此外,在异养细菌群落结构和功能研究方法上亦是有益的探讨。 相似文献
